Suzuki gave the GSX-R750 a full model change for 2000, continuing the model's long-running role as the company's 750-class supersport while cutting weight and refining the engine and chassis package. Suzuki's official GSX-R750 digital archive describes the 2000 model as the lightest GSX-R750 produced to that point, with a dry mass of 166 kg for the main-market specification. The redesign combined a more compact engine, lighter frame and longer swingarm with a short overall wheelbase to improve traction and handling.

The GSX-R750 uses a 749 cm³ liquid-cooled DOHC inline-four with a 72.0 × 46.0 mm bore and stroke and a 12.0:1 compression ratio. The 2000 factory specification documents electronic fuel injection, a wet multi-plate clutch and a six-speed constant-mesh transmission. Final drive is by chain with a 42/17 reduction ratio. Suzuki's archive notes that the 2000 redesign also adopted a double-valve fuel-injection arrangement in the throttle body, combining rider-operated and electronically controlled throttle valves to improve torque, power and combustion efficiency.

The chassis retained the aluminum twin-beam supersport layout but was substantially lightened and repackaged. Factory specifications list a 1,410 mm wheelbase, 830 mm seat height, inverted fully adjustable front suspension and a link-type rear suspension with adjustable preload, rebound and compression damping. Twin front disc brakes and 17-inch radial tires complete the road-racing-oriented package. These characteristics make the 2000-generation machine a fuel-injected 750cc supersport motorcycle focused on low mass, compact dimensions and high-rpm performance.
